Shoot cultures of Vitis vinifera (vine grape) different cultivars as a promising innovative cosmetic raw material – phytochemical profiling, antioxidant potential and whitening activity

Abstract
The primary purpose of this work was the initiation and optimization of shoot cultures 25 of different Vitis vinifera L. cultivars: cv. Chardonnay, cv. Hibernal, cv. Riesling, cv. Johanniter, 26 cv. Solaris, cv. Cabernet Cortis and cv. Regent. CThe cultures were maintained onover 30-days 27 growth cycles usingon two media: Murashige and Skoog (MS) and Schenk and Hildebrandt (SH) 28 with various concentrations of plant growth regulators. TThe tested media types used (W1 – 29 W4) contained varying concentrations of differed in 6-benzylaminopurine (BA) concentration 30 inas well as the aaddition toof indole-3-butyric acid (IBA) and 1-naphthaleneacetic acid 31 (NAA).were: ‘W1’ - MS, 0.9 mg/L 6-benzylaminopurine (BA) and 0.1 mg/L indole-3-butyric acid 32 (IBA), ‘W2’ - MS, 1.5 mg/L BA and 0.2 mg/L 1-naphthaleneacetic acid (NAA), ‘W3’ - SH, 0.9 mg/L 33 BA and 0.1 mg/L IBA and ‘W4’ - SH, 1.5 mg/L BA and 0.2 mg/L NAA. High performance liquid 34 chromatography coupled to mass spectrometry (UPLC-MS) was used for metabolomic profiling. 35 In all tested extracts, 45 compounds were identified (6 amino acids, 4 phenolic acids, 13 flavan-3- 36 ols, 3 flavonols and 19 stilbenoids). PThe principal component analysis (PCA) was performed 37 to assess the influence of genotype and medium on the metabolic content. PCA showed that met- 38 abolic contents wasere mainly influenced by genotype and to a lesser extent by medium compo- 39 sition. MS media variants induced the amino acids (e.g. L-leucine, L-phenylalanine), procya- 40 nidins (e.g. B1, B3) and flavan-3-ols (catechins) production. In addition, the antioxidant potential 41 and antityrosinase activity was measured spectrophotometrically. The studies of antioxidant ac- 42 tivity clearly revealed that the tested extracts, regardless of the culture media, showed very high 43 efficiency in reducing free radicals. The strongest tyrosinase inhibition capacity was showed for 44 shoots cv. Hibernal cultured on SH medium supplemented with NAA ‘W4’ with anthe inhibition 45 of 17.50%. Out of the tested V. vinifera cvs., the best results were indicated for cv. Hibernal. Studies 46 showedproved that V. vinifera cvs. in vitro cultures can be proposed as an alternative source of 47 plant material with a potential use in cosmetic industries.
