Novel pyridine derivatives as non-covalent fluorescent sensors for selective detection of bio-particles in medical diagnostics

Abstract
Background Albumin is a major serum protein that acts as the main transport protein in the body. It acts as a transporter of many essential compounds such as fatty acids, hormones, metal ions, and drugs. Therefore, it is important to search for convenient and simple methods to determine albumin content in the body. Thanks to advantages of fluorescent spectroscopy and fluorescence sensors it becomes possible. Aims In view of the importance of the development of new sensors, it was decided to study new 3-(2pyridyl)chromen-2-one derivatives. For this purpose, absorbance and fluorescence studies as well as the interaction of the obtained derivatives with BSA were carried out in order to determine the binding parameters and the limits of detection and quantification. Results The derivatives under study have a high molar extinction coefficient value of over 10,000 dm3·mol-1·cm-1 suggesting their high potential as fluorescent sensors. The probes tested bind to BSA and exhibit high sensitivity to changes in protein concentration in the system. The values of the detection and quantification limits are of the order of magnitude 10-7 and 10-8, while the concentration of albumin in blood is of the order of magnitude 10-5. The results showed that the quenching of BSA fluorescence occurs via a static fluorescence quenching mechanism. Obtained thermodynamic parameters such as negative Gibb’s free energy change, entropy and enthalpy change indicate that the binding of the probes to BSA is due to a spontaneous complexation. Moreover, the determined binding parameters of the interaction of the sensors with albumin indicate the possibility of using the studied derivatives for the role of non-covalent fluorescent sensors.
